Abstract
Purpose: Describe the parameters evaluated per FFPE human liver sample received/obtained for the University of Minnesota SenNet TMC. These parameters are used to evaluate whether the tissue can be characterized as "normal" vs "diseased" prior to further protocols.
Steps
Prepare two slides with serial sections of the FFPE liver sample -- 1 section per slide.
Section A -- Stain with H&E.
Section B -- Stain with trichrome.
Mount coverslip.
Schedule evaluation with pathologist (ideally with a strong liver background).
Request assessment and record of the following scores:
-
Steatosis, macro %
-
Steatosis, score (0, 1, 2, 3)
-
Ballooning (0, 1, 2)
-
Inflammation (0, 1, 2, 3)
-
Fibrosis (0, 1a, 1b, 1c, 2, 3, 4)
-
Any comments on preservation/quality of the sample
-
Other/Additional identifications (e.g. the presence of NASH or other disease states)
Record these values and consider them prior to moving the sample forward for experimentation processing.
Inventory and archive H&E and trichrome stains at room temperature -- Can be scanned if future images are required.
Inventory and archive FFPE blocks at room temperature.
