Steps

Procedure

1.

Inject mouse intraperitoneally with 250µg of Ultra-LEAF Purified anti-mouse CD4 or Rat IgG2b isotype control antibodies (Biolegend, Cat #100457 and Cat #400644; San Diego, CA) 3 and 1 days prior to immunization.

2.

After immunization, inject mice with 250µg of antibody weekly.

3.

Assess cell depletion by flow cytometric analysis of mouse blood serum.

4.

For depletion of CD8+ T cells, inject the mouse intraperitoneally with 250µg of Ultra-LEAF Purified CD8+ or Rat IgG2b isotype control antibodies (Biolegend Cat # 100746 and Cat #400644; San Diego, CA) 2 days prior to immunization.

5.

After immunization, inject mice with 250µg of each antibody 9- and 19-days post immunization.

6.

Assess cell depletion by flow cytometric analysis of mouse blood serum.
